Novo Nordisk: Mini-Robots and Protective Shields for the Stomach

Pharmaceutical giant Novo Nordisk relies on two extremely sophisticated technologies in its peptide therapies to outwit aggressive stomach acids. In the case of the oral semaglutide tablet Rybelsus, the co-formulated excipient salcaprozate sodium protects the sensitive peptide by neutralizing the acidic digestive enzyme pepsin and increasing the permeability of the stomach wall by 43%. With Amycretin, the company is already advancing its next promising candidate, which demonstrated significant weight loss of about 13% in initial trials. Together with MIT researchers, Novo Nordisk is even developing a mechanical injection capsule that autonomously and completely painlessly injects biologically active ingredients directly into the tissue layer of the stomach wall via a microneedle. In insulin trials, this mechanical approach achieves an astonishing bioavailability of up to 80%. This proves that smart delivery systems can make all the difference.

With its patented ENHANZE technology, the U.S. biotech company Halozyme Therapeutics dominates the field of enzymatic tissue penetration, replacing hours-long infusions via syringe. For fiscal year 2026, Halozyme Therapeutics is targeting total revenue of up to $1.81 billion, which would represent significant growth of 30%. A recently concluded partnership secures the company revenue-based royalties in the mid-single-digit percentage range for a novel antibody used to treat severe bone marrow diseases. However, its monopoly is faltering, as South Korean competitor Alteogen is also gaining market share with its own enzyme platform. In addition, Halozyme has already suffered significant legal setbacks this year in British and U.S. patent courts, resulting in the loss of key intellectual property rights.

BioNxt Solutions: A Breakthrough with Dissolvable Ultra-Thin Films

In the shadow of the pharmaceutical giants, the Canadian-German biotech company BioNxt Solutions is developing innovative sublingual dissolvable films designed to revolutionize the traditional administration of medications. The main focus is on the BNT23001 program, which embeds the proven active ingredient cladribine into a special hydrophilic polymer matrix for the treatment of multiple sclerosis. Since many patients suffer from severe swallowing difficulties, the dissolvable thin film under the tongue offers significant advantages. Pharmacokinetic studies in large-animal models demonstrate that the cladribine dissolvable film achieves more than 40% higher systemic bioavailability than the conventional reference tablet, Mavenclad. This superiority allows for a lower dose, thereby sparing patients’ immune systems and reducing costs. Further development of BNT23001 is progressing rapidly. Together with the Munich-based specialty laboratory Gen-Plus, BioNxt has successfully submitted clinical trial samples. BioNxt Solutions’ sublingual cladribine platform is protected by European unitary patent EP4539857 in 18 EU member states. This intellectual property right is supplemented by priority applications in the US, Canada, Japan, and Australia.

A turnaround is in the air at BioNxt.

In addition to its flagship project, BioNxt Solutions is strategically expanding its product portfolio to target other autoimmune diseases and longevity applications. The BNT23002 program is paving the way for the use of cladribine in myasthenia gravis, an autoimmune disease that causes muscle weakness, while BNT24002 and BNT24003 target the indications of lupus nephritis (kidney inflammation) and rheumatoid arthritis (chronic inflammation of the synovial membrane). With the BNT24005 program, BioNxt Solutions aims to replace the weight-loss injection and is researching an oral semaglutide film for the obesity market that does not infringe Novo Nordisk’s patents.

BioNxt: Diversified Portfolio – Even a Single Approach Can Make a Difference

Catalysts for future performance include clinical data and the continued development of the semaglutide dissolvable film in the multi-billion-dollar market for obesity therapies. With its various projects, BioNxt has a diversified portfolio and has several irons in the fire. The fact that developing therapies and biotechnology platforms is a challenging business is evident in the stock’s share price, which has been falling for months. It seems that the patience of many market participants is wearing thin. Given a market capitalization of only around CAD 38 million, however, this could also create opportunities—if a groundbreaking study succeeds, a trend reversal is likely. Speculative investors can keep an eye on BioNxt Solutions stock.
